Growth hormone secretagogue - GHRH analogue

CJC-1295

GHRH analogue in two forms that are not interchangeable

CJC-1295 is a synthetic analogue of growth hormone-releasing hormone (GHRH). It is studied for its effect on endogenous growth hormone secretion and, downstream, IGF-1.

The single most important thing to understand about this name is that it covers two different molecules. 'With DAC' carries a drug affinity complex that binds albumin and extends the half-life to roughly a week. 'Without DAC' - also called mod GRF 1-29 - has a half-life measured in minutes and is typically studied alongside a separate GHRP such as ipamorelin. Product listings frequently use the names loosely.

Mechanism

GHRH receptor agonism

Binds the GHRH receptor on pituitary somatotrophs, stimulating growth hormone release. This is the same receptor the body's own GHRH acts on.

Albumin binding (with DAC)

The DAC maleimide group attaches to serum albumin, which is what produces the week-long presence instead of a short pulse.

Pulsatility

Growth hormone is normally released in pulses. A long-acting analogue flattens that profile, which is a recognised research trade-off rather than an unqualified improvement.

Research status

Human studies of GHRH analogues are short-term and measure hormone levels rather than clinical outcomes. They show that GH and IGF-1 rise, which is mechanistically expected. What is not established is whether sustained elevation produces benefit or harm over time - and the long-acting profile has attracted specific attention on that point.

Verifying a batch

  • 01Establish which form a batch contains before anything else. With DAC and without DAC differ by a modification that mass spectrometry will show plainly.
  • 02For the without-DAC form, confirm the sequence is mod GRF 1-29 rather than unmodified GRF 1-29, which is degraded far faster.
  • 03Long-acting analogues place more weight on cold-chain handling.

Questions

What is the difference between CJC-1295 with DAC and without DAC?+

Half-life. With DAC binds albumin and persists for about a week; without DAC lasts minutes. They are different research tools, not two strengths of the same thing.

Is CJC-1295 the same as tesamorelin or sermorelin?+

No. All three are GHRH analogues but they are distinct molecules with different modifications and different evidence. Tesamorelin is the one with an approved human indication.

Why is it often combined with ipamorelin?+

GHRH and ghrelin-receptor agonists act on different receptors that converge on GH release, so pairing them amplifies the pulse in a way neither does alone. The pairing is studied; the long-term outcomes are not.
